in situ hybridization (ISH) is a technique that enables the localization of specific DNA or RNA sequences within tissue sections and cytology samples using labeled complementary strands. The target gene can be visualized through brightfield or fluorescence microscopy. Compared to IHC, which broadly localizes proteins within tissues, ISH offers superior sensitivity. ISH not only identifies cells positive for a specific DNA or RNA sequence but also quantifies their level of positivity within individual cells. This makes ISH particularly valuable for studies requiring precise gene-level data within a morphological context.

We offer all methods of probe detection to uncover the morphological data of your samples. Our ISH services include fluorescence in situ hybridization (FISH), silver-enhanced in situ hybridization (SISH), and chromogenic in situ hybridization (CISH). For studies requiring a more comprehensive view, we also offer multiplex assays that combine ISH with immunohistochemistry (IHC) on the same slide, enabling simultaneous gene and protein expression analysis in situ.

FISH assays are performed using the Abbott VP2000 with hybridization steps completed on calibrated ThermoBrite heating modules. SISH assays are run on the Benchmark XT and Ultra automated systems. Multiplex CISH with IHC is performed on the Leica Bond autostainer. FISH signal evaluation and quantification is conducted using Zeiss Axioplan microscopes paired with the BioView imaging system. Chromogenic ISH slides are captured using whole slide digital imaging. All equipment is extensively qualified and undergoes regular quality checks.

Yes. CellCarta has extensively evaluated RNAscope (ACD) nucleic acid in situ hybridization technology for mRNA detection in FFPE tissue. RNAscope is validated for use on FFPE cell preparations and can be requested for any target of interest. As each specimen type presents unique challenges, the technology will undergo custom validation to fine-tune the procedure to your specific samples. Client specimens are tested during an assay optimization phase, and results determine whether further validation steps are required.
