PRAS40 is activated by Akt, and phosphorylation of this protein allows it to bind to 14-3-3, a cytosolic anchor protein. PRAS40 also binds mTOR (mammalian target of rapamycin), a highly conserved PI3K protein that is also involved with cellular survival with 2 complexes: mTORC1 (rapamycin-sensitive) and mTORC2 (rapamycin-resistant). Inhibition of mTOR signaling stimulates PRAS40 binding to mTOR, which in turn inhibits mTOR activity.
