Phosphatase and tensin homolog deleted on chromosome ten (PTEN) is a phosphatase that is expressed in most normal cells but is frequently lost or downregulated in cancer. PTEN is a major negative regulator of the Akt/mammalian target of rapamycin (MTOR) pathway. Selection of patients based on loss of PTEN expression might help to predict the sensitivity of tumor cells to rapamycin analogues.
